چکیده
Coconut palms are highly susceptible to diseases caused by different pathogens, and replanting with resistant varieties is the best way to manage them. Obtaining a collection of resistance gene analogues (RGAs) is an effective strategy to identify genomic regions linked to disease resistance. We have successfully used a comparative genomics approach to amplify putative RGAs from the coconut root (wilt) disease resistant cultivar Chowghat Green Dwarf (CGD) by using primers designed based on conserved motifs of the NBS-LRR domain of the date palm. The amplifi ed sequences were cloned, sequenced and characterized. The coconut RGAs had high identity to monocot NBS-LRRs. A complete structural analysis and 3-D modeling of the NBS domain of coconut RGA was also undertaken. Real-time quantitative polymerase chain reaction analysis indicated that the isolated coconut NBS-LRR class RGAs was expressed more in root (wilt) disease resistant genotypes than in susceptible ones. This study would provide a base for future efforts to map disease resistant traits in coconut.

Study of new NBS-LRR genes analogues in cucurbits native types in Iran
Nucleotide binding site leucine-rich repeats (NBS-LRR) accounting for the main disease resistance proteins play an important role in plant defense against pathogen attack. The current study aimed to identify new NBS-LRR gene members in native types of cucurbit species in Iran. Accordingly, DNAs of melon, cucumber and cantaloupe native types to Iran were identified using three primer pairs. PCR ...
متن کاملIsolation and characterization of NBS–LRR resistance gene analogues from mango
The nucleotide-binding site (NBS)-leucine-rich repeat (LRR) gene family is a class of R genes in plants. NBS genes play a very important role in disease defence. To further study the variation and homology of mango NBS-LRR genes, 16 resistance gene analogues (RGAs) (GenBank accession number HM446507-22) were isolated from the polymerase chain reaction fragments and sequenced by using two degene...
متن کاملMolecular Cloning and Characterisation of a Non-tir-nbs-lrr Type Disease Resistance Gene Analogue from Sugarcane
DISEASE resistance gene cloning is one of the main targets in modern sugarcane research. In this study, several primers were designed according to the conserved motifs in the NBS regions of known resistance genes to amplify sequences from cDNA of sugarcane variety NCo376 by polymerase chain reaction (PCR). In total, six NBS-LRR type resistance gene analogs (RGAs) were cloned, for which the dedu...
متن کاملIdentification and characterization of a NBS–LRR class resistance gene analog in Pistacia atlantica subsp. Kurdica
P. atlantica subsp. Kurdica, with the local name of Baneh, is a wild medicinal plant which grows in Kurdistan, Iran. The identification of resistance gene analogs holds great promise for the development of resistant cultivars. A PCR approach with degenerate primers designed according to conserved NBS-LRR (nucleotide binding site-leucine rich repeat) regions of known disease-resistance (R) gene...
متن کاملThe CC-NBS-LRR Subfamily in Pinus monticola: Targeted Identification, Gene Expression, and Genetic Linkage with Resistance to Cronartium ribicola.
ABSTRACT To investigate disease resistance gene analogs (RGAs) encoding coiled-coil-nucelotide-binding site-leucine-rich repeats (CC-NBS-LRR) proteins in western white pine, degenerate primers targeting the conserved motifs in the NBS domain were designed to amplify RGAs from genomic DNA and cDNA. Sixty-one distinct RGAs were identified with identities to well-known R proteins of the CC-NBS-LRR...
